I don't understand what "die" is doing here, I assume that it refers back to "meinung" in the previous phrase. Could it be left out or is it necessary?

Hi sfpugh, 

the "die" is actually an article for "Kenntnisse und Qualifikationen".
The attribute "für Ihre Stelle erforderlichen" in between article and nouns functions as closer information about the nouns "Kenntnisse und Qualifikationen" - "erforderlichen" is an adjective.

Thank you for pointing that out,  I was confused by "für Ihre Stelle" as that would go after the nouns in English. It certainly wouldn't go where it is here.
I suppose you could put "for your position" at the start of the phrase for emphasis "I am definitely of the opinion that, for your position,  I have the necessary skills and qualifications." but it sounds a bit odd.
